为了研究在基于纳米金的肿瘤光动力学疗法以及肿瘤热疗中纳米金在细胞中的分布及代谢
利用多光子荧光显微镜技术对在热疗中纳米金结合的肿瘤细胞进行了多次扫描成像
并采用一种基于区域的活动轮廓模型图像分割算法
对与纳米金结合的细胞的多光子荧光显微图像进行处理
在一幅具有多个细胞的图像中分割出单个细胞的轮廓。根据细胞断层切片图像的轮廓采用移动立方体算法
对细胞进行三维重建
绘制出细胞表面的三维模型
再利用纳米金的荧光寿命和光强来标定纳米金在细胞具体位置的分布。结果表明
在一定时间内
纳米金会进入细胞并在不同的区域内呈现一定的分布。这与抗原抗体结合会导致抗体的内化
从而使纳米金进入细胞内的事实相符。这种技术将会为更加直观、全面地研究纳米金与细胞的相互作用以及纳米金在细胞内具体位置(比如某个细胞器内的分布)提供一种可行的方法。
To analyze the distribution and metabolism of gold nanoparticles in cells during the gold nanoparticles-based photodynamic therapy and photo-thermal therapy
the multiphoton fluorescence microscopy was used to image the gold nanoparticles labeled cells
and a region-based active contour model image segmentation algorithm was chosen to process the multiphoton fluorescence images of gold nanoparticles labeled cell. 60 thin slice images were reconstructed for a 3D cell image to draw a three-dimensional model of cell surface. The locations of gold nanoparticles in or on the cell were marked according to the fluorescence lifetime and intensity of gold nanoparticles. The results show that the gold nanoparticles can get into the cells
which is in accordance with the fact that antibody is internalized when the antibody combines with antigen. This technology is expected to more comprehensively investigate the interaction between gold nanoparticles and cell as well as the specific distribution of gold nanoparticles

in a certain cell organelle.
