An amperometric immunosensor for human chorionic gonadotrophin(HCG)in the saliva is developed based on HCG antibody(anti-HCG)coated mercapto succinic amide(HL)/gold nanoparticles(AuNPs)multilayer membrane modified Au electrode. HL/AuNPs monolayers are modified alternatively and form multiple membranes through Au-NH
2
and Au-S bonds by layer-by-layer self-assembly technology. K

Fe(CN)

is employed as electron probe for detection. The antigen-antibody immune complex formed on the electrode surface is able to hinder electron transport
so the redox current of K
3
Fe(CN)
6
is decreased. The decreased value of ΔI
p
is related to the extent of immune-reaction on membrane surface
which serves as the quantitative basis of HCG content. The experimental results show that multilayer AuNPs membrane can adsorb more anti-HCG
and obviously broaden the linear detection range of HCG to 0.1-100 mg/L with detection limit of 0.056 mg/L(3σ).
